VirusTotal rating: 4.2 / 5 (A) .The plasma membrane protein Hv1p of Saccharomyces cerevisiae belongs to a family of small tetra-span membrane proteins (Bae and Cordes, EMBO J. 8, 693-701, 1989). The yeast homologues of Hv1p, such as Ypk1p (Newstead, et al., Science, 273, 1648-1652, 1996), Ypk2p (Newstead, et al., Science, 273, 1648-1652, 1996), Ypk3p (Christie, et al., Science, 274, 1895-1898, 1996), and Hkr1p (Hayakawa, et al., Science, 272, 755-758, 1996), are all localized at the plasma membrane. The N-terminal half of Ypk3p, which is highly homologous to Hv1p, is responsible for plasma membrane localization. In contrast, the C-terminal half of Ypk3p, which is not homologous to Hv1p, is essential for DNA damage responses. The N-terminal half of Ypk3p localizes at the plasma membrane and is required for the activation of the G2/M checkpoint by DNA damage (Christie, et al., Science, 274, 1895-1898, 1996). On the other hand, the C-terminal half of Ypk3p is localized in the nucleus, but is not required for DNA damage responses. Thus, Ypk3p can function as a dual-site protein. The protein domain responsible for the dual-site function of Ypk3p has not been identified. As shown in FIG. 1, a schematic diagram showing the domain structure of Ypk3p (Christie, et al., Science, 274, 1895-1898, 1996), Ypk3p contains a N-terminal domain (Ypk3p-(a)) with two pleckstrin homology (PH) domains, a central domain (Ypk3p-(b)) with a SH3-like domain and a pleckstrin homology (PH) domain, and a C-terminal domain (Ypk3p-(c)).
